Label-free visualization of nano-thick biomolecular binding by electric-double-layer modulation
We developed a label-free and highly surface-sensitive bioimaging technique, named as electric-double-layer modulation imaging (EDLMI), which utilized the phenomenon that charge density of electric double layer, accumulated at the interface between a semiconductor and electrolyte solution, responded sensitively to biomolecular adsorption. The phenomenon was visualized by lock-in imaging in synchronization with the electric-double-layer modulation, allowing to monitor nano-thick biomolecular adsorption in real time with a wide-range magnification unlike conventional label-free bioimaging techniques. We demonstrate that EDLMI can differentiate specific and non-specific binding between antibodies.
